As a child, Henderson began performing at local clubs during charity events and various holidays. After school, Shirley also attended a theater club. After graduating from college in Fife when she was seventeen, she moved to London, where she studied acting for three years at Hydehall School.
For the first time on the screens Shirley appeared in the children's television movie "Shadow of the Stone". For a long time after that, she played only in the theater, and returned to the movies only in the 90s, playing a small role in the military series Wish Me Good Luck. Since then, she has combined work in television and the London National Theatre.
